چکیده

Cholesterol side chain cleavage has been shown to be dependent on the electron carrier chain involving a cytochrome P-450 reductase system, flavoprotein and nonheme iron-protein of the ferredoxin type, and the oxygen-activating cytochrome P-450. The reaction is markedly inhibited by carbon monoxide. The cytochrome oxidase system inhibits the cleavage reaction by diverting the flow of electrons through the cytochrome oxidase pathway. Inhibition is reversed by cyanide. Steroid products resulting from the cleavage reaction are pregnenolone, progesterone, 11 -deoxycorticosterone, and corticosterone. These products were isolated and identified by paper chromatographic techniques and recrystallization to radiochemical homogeneity.

Inhibition of cholesterol side chain cleavage by active site directed antibody to corpus luteum cytochrome P-450.

Goat antibody IgG produced against bovine corpus luteum mitochondrial cytochrome P-450 (P-450scc) associated with cholesterol side chain cleavage (CSCC) was used to compare immunological characteristics of mitochondrial cytochrome P-450s from the bovine adrenal cortex (BAM), bovine corpus luteum (BCLM), and human placenta (HPM). In Ouchterlony double diffusion, anti-P450scc produced a single ba...

Evidence for a higher molecular weight precursor of cholesterol side-chain-cleavage cytochrome P-450 and induction of mitochondrial and cytosolic proteins by corticotropin in adult bovine adrenal cells.

Adult bovine adrenal cortical cells in monolayer culture were used to study the induction of cholesterol side-chain-cleavage cytochrome P-450 by corticotropin (ACTH). In the presence of 1 microM ACTH, there was a 4-fold increase in cortisol production by these cells over a 72-hr period and a corresponding increase in total cytochrome P-450 content. The incorporation of [35S]methionine into a nu...

Induction of synthesis of cholesterol side chain cleavage cytochrome P-450 by adrenocorticotropin in cultured bovine adrenocortical cells.

The long-term action of adrenocorticotropin (ACTH) to stimulate side chain cleavage of cholesterol has been studied utilizing confluent monolayer cultures of adult bovine adrenal cortex cells maintained for periods of time up to 72 h in the absence or presence of ACTH (lo-’ M). Following incorporation of [36S]methionine into cellular protein at various times after ACTH addition, cholesterol sid...

Functional reconstruction of bovine P450scc steroidogenic system in Escherichia coli

Mammalian cytochrome P450scc enzyme system catalyzes the initial step in steroid hormone biosynthesis—cholesterol hydroxylation followed by cleavage of the side-chain to yield pregnenolone. This system consists of three components—the cytochrome P450scc (CYP11A1), a flavoprotein (NADPH-adrenodoxin reductase, AdR) and an iron-sulfur protein (adrenodoxin, Adx). In this work, the three-component e...

Common characteristics of the cytochrome P-450 system involved in 18- and 11 beta-hydroxylation of deoxycorticosterone in rat adrenals.

18- and 11beta-Hydroxylation of deoxycorticosterone and side chain cleavage of cholesterol were studied in mitochondria and submitochondrial reconstituted systems prepared from rat and bovine adrenals. A mass fragmentographic technique was used that allows determination of hydroxylation of both exogenous and endogenous cholesterol. The following results were obtained. (1) Treatment of rats with...
